摘要:
目的:以蒸发光散射检测器(ELSD)测定党参中苍术内酯Ⅲ的含量,并与二级管阵列检测器(PAD)测定结果进行比较。方法:采用RP-HPLC法,Hypersil ODS-2柱(5 μm,4.6 mm×250 mm),甲醇-水为流动相(67:33),流速1.0 mL/min, 柱温30℃。结果:用ELSD检测党参药材中苍术内酯Ⅲ的回归方程为A=1.30×104C-2.33×104, 相关系数r=0.9938, 平均回收率为103.65%, RSD为 0.93%。结论:以ELSD测得的党参药材中苍术内脂Ⅲ的含量与PAD测得的党参药材中苍术内脂Ⅲ的含量基本一致,以 ELSD-RP-HPLC法测定党参药材中苍术内脂Ⅲ的含量的方法简便、易行、准确、可靠。
Abstract:
Objective: To determinate the content of atractylenoide Ⅲ in Dangshen by ELSD and PAD,and compare the results. Methods: Adopt RP-high performance liquid chromatography (HPLC) and Hypersil ODS-2 column (5 μm,4.6 mm×250 mm).Mobile phase was methanol-water(67:33), flow-rate was 1.0mL/min, column temperature was 30℃. Results: Linear equation of atractylenolide Ⅲ in Dangshen by ELSD was A=1.30×104C-2.33×104.Relative coefficient was 0.9938. The average recovery was 103.65 percent and relative standard deviation was 0.93 percent. Conclusion: The content of atractylenolide Ⅲ by ELSD and PAD was unanimous basically. The method by ELSD was simple, reliable, accurate and practical.
