摘要:
目的 比较西藏不同产区长梗秦艽中龙胆苦苷和马钱苷酸含量的差异。方法 收集西藏3个地区共8份长梗秦艽样品,采用高效液相色谱(HPLC)法对其中龙胆苦苷和马钱苷酸的含量进行测定。色谱柱为Kromasil C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为甲醇-0.1%磷酸水溶液,梯度洗脱,流速为1 mL/min,检测波长245 nm,柱温35℃,进样量10 μL。结果 龙胆苦苷回归方程为Y=1 050X+34.6,r=0.999 7,结果表明龙胆苦苷在61.063~977.000 μg/mL范围内线性关系良好;马钱苷酸回归方程为Y=898.7X+47.9,r=0.999 9,结果表明马钱苷酸在11.563~185.000 μg/mL范围内线性关系良好。龙胆苦苷含量均值依次为拉萨 > 林芝 > 昌都,马钱苷酸含量均值依次为拉萨 > 昌都 > 林芝;且西藏8个产地中只有昌都地区一个样地的长梗秦艽中龙胆苦苷与马钱苷酸的含量总和未达到2015版《中华人民共和国药典》标准(2.5%)。结论 该方法快捷、有效,可用于长梗秦艽中龙胆苦苷与马钱苷酸的含量测定。
Abstract:
Objective To compare the difference of component contents of Gentiana waltonii from different areas in Tibet. Methods Eight samples of Gentiana waltonii were collected from 3 regions in Tibet. The contents of gentiopicroside and loganin acid were determined by HPLC. The samples were separated on the column of Kromasil C18 (4.6 mm×250 mm,5 μm),which moving phase was methanol-0.1% phosphoric acid solution as well as gradient elution was used. The flow rate was 1 mL/min. The detected wave-length was at 245 nm. The column temperature was at 35℃ and the sample size was 10 μL. Results The regression equation of gentiopicroside was Y=1 050X+34.6,r=0.999 7,showing a good linearity in the range of 61.063-977.000 μg/mL; the regression equation of loganic acid was Y=898.7X+47.9,r=0.999 9,showing a good linearity in the range of 11.563-185.000 μg/mL. The contents of gentiopicroside in three regions showed as Lhasa > Nyingchi > Chamdo and that of loganic acid showed as Lhasa > Chamdo > Nyingchi. Among the 8 producing areas in Tibet,the total content of gentiopicroside and loganin acid in Gentiana waltonii only from Chamdo was under 2.5% (specified standard in Chinese Pharmacopoeia). Conclusion This method is simple and accurate,which can be used to determine the gentiopicroside and loganic acid in Gentiana waltonii.
