摘要:
目的 建立高效液相色谱(HPLC)法测定安徽产石菖蒲中α-细辛醚和β-细辛醚含量的方法,并对安徽产石菖蒲药材进行质量评价。方法 采用VP-ODS(4.6 mm×150 mm,5 μm)色谱柱,对以下色谱条件进行考察,流动相分别为甲醇-水(含0.1%甲酸)60:40,55:45,70:30,温度分别为35,30,25℃,检测波长分别为257,253,250 nm,确定最佳色谱条件,并在最佳色谱条件下测定安徽产石菖蒲中α-细辛醚和β-细辛醚的含量。结果 最佳色谱条件:流动相为甲醇-水(含0.1%甲酸)55:45,流速1.0 mL/min,柱温30℃,检测波长250 nm。α-细辛醚平均回收率为99.17%,RSD为2.35%;β-细辛醚平均回收率为99.53%,RSD为2.14%。结论 该方法灵敏简便、重复性良好、结果准确可靠,可用于石菖蒲的质量评价。
Abstract:
Objective To establish content test methods for α-asarone and β-asarone in Acorus tatarinowii Schott and make medicinal material quality assessment for it. Methods The chromatographic conditions were observed on the column of VP-ODS(4.6 mm×150 mm,5 μm),which moving phases were methanol and water (including 0.1% formic acid) 60:40,55:45,70:30,column temperature was at 35,30,25℃ and detected wave-length was at 257,253,250 nm. The best conditions were taken to determine the content of α-asarone and β-asarone in Acorus tatarinowii Schott. Results The best chromatographic conditions were as following:moving phase was methanol and water (55:45); flow rate was 1.0 mL/min; column temperature was at 30℃ and detected wave-length was at 250 nm. The average recovery rate of α-asarone and β-asarone were 99.17% and 99.53% respectively,while RSD were 2.35% and 2.14%. Conclusion The method is simple,sensitive and with good reproducibility,high accuracy,which can be used to evaluate the quality of Acorus tatarinowii Schott.
