摘要:
目的 建立中风康复胶囊中丹参药材的定性、定量检测方法,并制定质量标准。方法 采用薄层色谱(TLC)法,以丹参酮ⅡA为对照品,三氯甲烷-甲苯-乙酸乙酯-甲醇-甲酸(6∶4∶8∶1∶4)为展开剂,建立其定性鉴别方法;采用高效液相色谱(HPLC)法,以丹参酮ⅡA为对照品,色谱柱为Diamonsil C18(5 μm,4.6 mm×200 mm),流动相为乙腈-0.02%磷酸水溶液(75∶25),流速1.0 mL/min,检测波长270 nm,柱温25 ℃,测定中风康复胶囊中丹参酮ⅡA的含量。结果 所建立TLC法斑点清晰,分离效果好。含量测定中丹参酮ⅡA的回归方程为Y=2 980X-20.286(r=0.999 4),说明丹参酮ⅡA在0.035 6~0.605 0 mg/mL范围内呈良好的线性关系;平均回收率为100.00%,RSD为0.97%。结论 本法简便可行,重复性好,可用于中风康复胶囊中丹参药材的质量控制。
Abstract:
Objective To establish the qualitative and quantitative test method of salvia miltiorrhiza in Zhongfeng Kangfu Capsule,and draw up quality standards.Methods Qualitative way to distinguish was made to test salvia miltiorrhiza in Zhongfeng Kangfu Capsule by thin layer chromatography (TLC) method;tanshinone ⅡA was used as a control and chloroform-toluene-ethyl acetate-methanol-formic acid (6:4:8:1:4) was used as the developing solvent.High-performance liquid chromatography (HPLC) was used to determine the content of salvia miltiorrhiza in Zhongfeng Kangfu Capsule.Tanshinone ⅡA was used as the control.The chromatographic column was Diamonsil C18(5 μm,4.6 mm×200 mm);the mobile phase was acetonitrile-0.02% aqueous phosphoric acid (75:25),the flow rate was 1.0 mL/min,the detection wavelength was 270 nm,and the column temperature was 25℃. Results The spots were clear and separating effect was good.The regression equation of Tanshinone ⅡA was Y=2 980X-20.286(r=0.999 4),showing a good linearity in the range of 0.035 6 to 0.605 0 mg/mL. The average recovery rate was 100.00% and the RSD was 0.97%.Conclusion This method is simple,effective with good repeatability and can be used for quality control of salvia miltiorrhiza in Zhongfeng Kangfu Capsule.
