摘要:
目的 建立运用反相高效液相色谱法测定甘草内生菌代谢物中甘草酸铵、甘草苷及甘草素含量的方法。方法 采用Phecda C18色谱柱(4.6 mm×250.0 mm,5 μm),柱温35℃,以乙腈-0.5%磷酸水溶液为流动相,梯度洗脱;检测波长为250 nm,流速1 mL/min,进样量10 μL。结果 甘草酸铵、甘草苷及甘草素的线性范围分别为0.013 0~0.325 5 μg(r=1.000 0),0.002 8~0.070 3 μg(r=0.999 5),0.002 2~0.055 2 μg(r=1.000 0),平均加样回收率分别为97.39%(RSD=1.24%),97.44%(RSD=1.75%),95.83%(RSD=1.79%)。结论 甘草内生菌代谢物中含有与宿主甘草相同的甘草酸铵、甘草苷及甘草素,本法操作简便、快捷,结果准确,可用于甘草内生菌代谢物中甘草酸铵、甘草苷及甘草素的含量测定。
Abstract:
Objective To establish content test methods for ammonium glycyrrhetate,liquiritin and liquiritigenin in the fermentation products of endophytes from Glycyrrhiza uralensis. Methods Phecda C18 (4.6 mm×250.0 mm,5 μm) chromatographic column was adopted,acetonitrile-0.5% phosphoric acid water was used as moving phase,and gradient elution was performed. With a wavelength of 250 nm,a velocity of 1 mL/min and an injection amount of 10 μL. Results The linearity ranges of ammonium glycyrrhetate,liquiritin and liquiritigenin were 0.013 0 to 0.325 5 μg (r=1.000 0),0.002 8 to 0.070 3 μg (r=0.999 5),and 0.002 2 to 0.055 2 μg(r=1.000 0). The average recovery rate was 97.39% (RSD=1.24%),97.44%(RSD=1.75%) and 95.83% (RSD=1.79%) respectively. Conclusion The metabolites of endophytes and host plant Glycyrrhiza uralensis have similar ammonium glycyrrhetate,liquiritin and liquiritigenin.The method is simple,fast and the results are accurate and can be used for content test for ammonium glycyrrhetate,liquiritin and liquiritigenin in the fermentation products of endophytes from Glycyrrhiza uralensis.
