摘要:
目的 研究肉苁蓉枸杞咀嚼片的质量控制方法。方法 采用薄层色谱(TLC)法对肉苁蓉枸杞咀嚼片中的肉苁蓉、枸杞子进行定性鉴别;采用高效液相色谱(HPLC)法对肉苁蓉主要成分松果菊苷、毛蕊花糖苷进行定量分析,色谱柱为Hypersil BDS C18(250.0 mm×4.6 mm,5 μm),流动相为甲醇-0.1%甲酸溶液,流速1 mL/min,检测波长330 nm,柱温25℃,进样量20 μL。结果 肉苁蓉和枸杞子的TLC特征斑点清晰、专属性强;松果菊苷和毛蕊花糖苷分别在41.8~418.0 μg/mL(r=0.999 9)和40~400 μg/mL(r=0.999 9)范围内线性关系良好,回收率分别为99.58%(RSD=0.85%)、99.31%(RSD=1.13%)。结论 所建立的定性和定量分析方法能有效控制该咀嚼片的质量。
Abstract:
Objective To establish the quality control methods of Roucongrong Gouqi Jujuepian.Methods TLC was used to identify Roucongrong (Cistanches Herba) and Gouqizi(Lycii Fructus).HPLC was used to determine the contents of echinacoside and verbascoside.The HPLC analysis was performed using a Hypersil BDS C18 (250.0 mm×4.6 mm,5 μm) column at 25℃.An isocratic elution with methanol and 0.1% formic acid was used at a flow rate of 1 mL/min.An aliquot of 20 μL of sample solution was injected onto the column and the detection wavelength was 330 nm. Results TLC spots features of Roucongrong (Cistanches Herba) and Gouqizi(Lycii Fructus) were clear with strong specificity.The linear ranges of echinacoside and verbascoside were 41.8 to 418.0 μg/mL (r=0.999 9) and 40 to 400 μg/mL (r=0.999 9).The average sample recoveries were 99.58% (RSD=0.85%) and 99.31% (RSD=1.13%).Conclusion The methods reported in this paper can be used scientifically and effectivelly to evaluate the quality of Roucongrong Gouqi Jujuepian.
