摘要:
目的 建立健脾瘦身丸的定性与定量检测方法,并制定质量标准。方法 对健脾瘦身丸中的决明子和山楂采用薄层色谱(TLC)法进行定性鉴别,对决明子中的有效成分大黄酚采用高效液相色谱(HPLC)法进行含量测定,色谱柱为Inertsil ODS-SP(4.6 mm×150.0 mm,5 μm),流动相为乙腈-0.1%磷酸溶液(60:40),检测波长430 nm,流速1.0 mL/min,柱温30℃,进样量10 μL。结果 所建立的TLC法斑点清晰,分离效果好,且阴性无干扰。含量测定中大黄酚的回归方程为Y=4.621 74×10-5X-0.023 729,r=0.999 9,表明大黄酚在0.045~0.680 μg范围内呈良好的线性关系。计算得平均加样回收率为97.20%,RSD为0.97%。结论 本研究建立的健脾瘦身丸的定性与定量检测方法操作简便、专属性强,结果准确可靠,可用于健脾瘦身丸的质量控制。
Abstract:
Objective To establish the qualitative and quantitative determination methods and set quality standard for Jianpishoushen Pills.Methods TLC was used to qualitative identification of Shanzha(Crataegi fructus) and Juemingzi(Cassina semen) in Jianpishoushen Pills;HPLC was used to content determination of chrysophanol in Cassina semen;Inertsil ODS-SP(4.6 mm×150.0 mm,5 μm)chromatography column was used with the mobile phase of acetonitrile-0.1% phosphoric acid (60:40),determine wavelength was 430 nm,the flow rate was 1.0 mL/min,the column temperature was 30℃,and sample size was 10 μL.Results The TLC spots were fairly clear and the result of separation was good.Moreover,the negative control showed no interference.The regression equation of content determination of Chrysophanol was Y=4.621 74×10-5X-0.023 729,r=0.999 9,which showed that there was a good linear relationship at the range of 0.045 to 0.680 μg (r=0.999 99) for chrysophanol;the average recovery was 97.2% and RSD was 0.97%.Conclusion The qualitative and quantitative method established in this study is simple,specific,accurate and reliable,which can be used for the quality control.
