摘要:
目的 观察藏药二十五味儿茶凝胶对慢性湿疹模型大鼠皮损及血清白细胞介素-4(IL-4)、γ-干扰素(IFN-γ)含量的影响。方法 将60只Wistar大鼠随机分为空白组,模型组,阳性对照组,藏药二十五味儿茶凝胶低、中、高剂量组(简称“凝胶低、中、高剂量组”)共6组,每组10只。采用2,4-二硝基氯苯复制慢性湿疹大鼠模型,各治疗组均经皮外涂相应药物,比较各组大鼠的皮损评分及血清IL-4、IFN-γ的含量。结果 与模型组比较,各治疗组大鼠皮损评分及血清IL-4含量均明显降低,INF-γ含量均明显升高,差异均有统计学意义(P<0.05或P<0.01);各治疗组之间比较,凝胶中、高剂量组大鼠皮损评分及血清IL-4含量均明显低于阳性对照组和凝胶低剂量组,INF-γ含量均明显高于阳性对照组和凝胶低剂量组,差异均有统计学意义(P<0.05或P<0.01)。结论 藏药二十五味儿茶凝胶能明显改善慢性湿疹模型大鼠皮损,降低大鼠血清IL-4含量,升高IFN-γ含量,其治疗慢性湿疹的作用机制与调节Th1、Th2型细胞因子有关。
Abstract:
Objective To observe the effects of Catechu Gel of Twenty-five Ingredients of Tibetan Medicine on skin lesions and the serum levels of IL-4 and IFN-γ in rats model of chronic eczema. Methods Sixty Wistar rats were randomly divided into 6 groups, including blank group, model group, positive control group, and the low, me-dium and high dose Catechu Gel of Twenty-five Ingredients of Tibetan Medicine groups (hereinafter referred to as Gel group), with 10 rats in each group. We used 2, 4-dinitrochlorobenzene to replicate rats model of chronic eczema. Each treatment group was smeared with the corresponding medicine on the outer skin, the skin lesions score and the serum levels of IL-4 and IFN-γ in rats of each group were compared. Results Compared with the model group, the skin lesions score and the serum level of IL-4 in rats of each treatment group all decreased significantly, the se-rum level of IFN-γ all increased significantly, there were statistically significant differences (P<0.05 or P<0.01). Compared among the treatment groups, the skin lesions score and the serum level of IL-4 in rats of the medium and high dose Gel groups were significantly lower than those of the positive control group and the low dose Gel group, the serum level of IFN-γ were significantly higher than that of the positive control group and the low dose Gel group, the differences were statistically significant (P<0.05 or P<0.01). Conclusion Catechu Gel of Twenty-five Ingredients of Tibetan Medicine can significantly improve the skin lesions in rats model of chronic eczema, decrease the serum level of IL-4 in rats, and increase the serum level of IFN-γ. Its mechanism in treating chronic eczema is related to the regulation of Th1 and Th2 cytokines.
