摘要:
目的 观察术芍配伍煎剂对慢传输性便秘模型大鼠肠功能及5-羟色胺(5-HT)、降钙素基因相关肽(CGRP)、乙酰胆碱酯酶(AchE)的影响。方法 将雌性Wistar大鼠随机分为空白组、模型组及术芍配伍煎剂组,每组10只。除空白组外,其余2组复制慢传输性便秘模型。造模成功后,空白组及模型组予0.9%生理盐水灌胃,术芍配伍煎剂组以6 g/kg剂量灌胃术芍配伍煎液,连续灌胃14 d。记录各组大鼠给药第2周的粪便粒数,测定粪便含水率,检测各组大鼠血清5-HT含量、结肠组织中CGRP含量及AchE活性。结果 与空白组比较,模型组大鼠治疗第2周的粪便粒数明显减少,粪便含水率明显降低,血清5-HT含量、结肠组织中CGRP含量及AchE活性明显降低,2组比较差异均有统计学意义(P<0.05);与模型组比较,术芍配伍煎剂组大鼠治疗第2周的粪便粒数明显增多,粪便含水率明显升高,血清5-HT含量、结肠组织中CGRP含量及AchE活性明显升高,2组比较差异均有统计学意义(P<0.05)。结论 术芍配伍煎剂可以通过调节慢传输性便秘模型大鼠的神经递质,恢复神经递质的正常释放,从而促进胃肠道的分泌和蠕动,恢复正常的排便功能。
Abstract:
Objective To observe the effects of Zhushao Peiwu Jianji(术芍配伍煎剂,ZPJ) on intestinal function and 5-hydroxytryptamine(5-HT),calcitonin generelated peptide(CGRP),acetylcholinesterase(AchE) of slow transit constipation in rats model. Methods Wistar female rats were randomly divided into blank group,model group and ZPJ group,with 10 rats in each.Except the blank group,the rest 2 groups duplicated the slow transit constipation models.After successful modeling,the blank group and model group were given 0.9% normal saline by gavage,the ZPJ group was given 6 g/kg dose of Zhushao Peiwu decoction by gavage,continuous administration by gavage for 14 days.The dejection amounts of the rats in each group at the 2nd week of administration were recorded,the fecal moisture content was determined,the serum content of 5-HT,the content of CGRP and activity of AchE in colonic tissues of the rats in each group was determined.Results Compared with the blank group,the dejection amounts of the rats in the model group at the 2nd week of administration decreased significantly,the fecal moisture content decreased significantly,the serum content of 5-HT,the content of CGRP and the activity of AchE in colonic tissues decreased significantly,there were statistically significant differences between the two groups(P<0.05).Compared with the model group,the dejection amounts of the rats in the ZPJ group at the 2nd week of administration increased significantly,the fecal moisture content increased significantly,the serum content of 5-HT and the content of CGRP and the activity of AchE in the colonic tissues increased significantly,there were statistically significant differences between the two groups(P<0.05).Conclusion ZPJ can regulate the neurotransmitters of rats with slow transit constipation,restore the normal release of neurotransmitters,thus promote the secretion and peristalsis of gastrointestinal tract,and restore the normal defecation function.
