摘要:
目的 建立通腑养髓汤高效液相色谱(HPLC)指纹图谱和主要成分含量测定方法。方法 采用HPLC法测定通腑养髓汤中芍药苷和盐酸小檗碱的含量,色谱柱为 KR100-5-C18柱(250.0 mm×4.6 mm,5 μm),流动相为甲醇(A)-0.1%磷酸溶液(B,0.03 mmol/L KH2PO4),梯度洗脱,流速1.0 mL/min,检测波长254 nm,柱温28 ℃。利用中药色谱指纹图谱相似度评价系统(2004A版)对色谱图进行分析,并测定主要成分芍药苷和盐酸小檗碱的含量。结果 建立了通腑养髓汤的特征指纹图谱,并指认出了12个共有峰中的2个主要共有峰芍药苷和盐酸小檗碱;芍药苷和盐酸小檗碱线性范围分别为125~4 000 μg/mL(r=0.999 9)、18.75~600.00 μg/mL(r=0.999 9),平均回收率分别为96.96%,98.54%,RSD分别为2.29%,1.42%。结论 建立的指纹图谱及含量测定方法可为通腑养髓汤的质量控制和评价提供参考。
Abstract:
Objective To establish the methods of HPLC fingerprint identification and content determination of principle components in Tongfu Yangsui Tang(通腑养髓汤). Methods HPLC method was used to determine the content of paeoniflorin and berberine hydrochloride in Tongfu Yangsui Tang. KR100-5-C18(250.0 mm×4.6 mm,5 μm) chromatography column was used with the mobile phase of methanol(A)-0.1% phosphoric acid(B,0.03 mmol/L KH2PO4) by gradient elution. The flow rate was 1.0 mL/min,the detection wavelength was 254 nm and the column temperature was 28 ℃. The similarity evaluation system for chromatographic fingerprint of TCM(2004A edition) was used to analyze the chromatogram and to determine the content of principle components,including paeoniflorin and berberine hydrochloride. Results The characteristic fingerprint of Tongfu Yangsui Tang had been established,and 2 of the 12 common peaks were identified as paeoniflorin and berberine hydrochloride. The linear ranges of paeoniflorin and berberine hydrochloride were 125 to 4 000 μg/mL (r=0.999 9) and 18.75 to 600.00 μg/mL (r=0.999 9),respectively. The average recovery was 96.96% and 98.54%,respectively,and the RSD was 2.29% and 1.42%,respectively.Conclusion The established fingerprint and content determination method can provide a reference for the quality control and evaluation of Tongfu Yangsui Tang.
